ALITA is a dual-gate design: Protease AND (Hypoxia OR ROS). In healthy tissues the gate stays OFF; in tumors it co-releases a debulking payload plus a local innate-immune stimulus—aiming for strong efficacy with fewer systemic side-effects.
How it works (high-level)
Our dual-gate activation ensures the therapy remains OFF in circulation and activates only within the tumor microenvironment.
Gate 1: Tumor-associated protease
The first gate uses proteases enriched in many tumors and largely quiescent in healthy tissues—providing the first layer of selectivity.
The construct is engineered for plasma stability until it encounters these enzymes.
Gate 2: Hypoxia or elevated ROS
The second gate requires either hypoxic conditions or elevated reactive oxygen species—common in solid tumors, uncommon in healthy tissue.
Requiring two distinct cues produces a stringent ON-state and minimizes off-tumor activation.
